Make a solution of 30 % (w/v) glycerol (autoclave)
Add 1 ml of late-log or early-stationary phase culture to 1 ml of glycerol solution
Mix
Freeze on dry ice
Store at -70°C
Strain storage (Lab. Hegemann)
Prepare an overnight culture in 10 ml medium (YPD or SD)
Harvest cells (4000 rpm – 5 min)
Resuspend pellet in 1 ml YPD + 1,5 ml sterile glycerol (85%)
Store on ice for at least 1 h
Flash freeze aliquots (1,25 ml) on dry ice
Store at –70°C
