Preparation of protein extracts from yeast

Protocol

  1. Collect yeast cells from a 2 x 1 ml culture by centrifugation in 1.5 ml microcentrifuge tubes (1 min at 6000 rpm in microcentrifuge)
  2. Remove supernatant
    Resuspend cell pellet in 600 ml TNE
    Add1.2 g of glass beads
  3. Mix cell suspension with the MM2000 mixer mill (Retsch, Haan, Germany) at highest oscillation frequency for 6 minutes.
  4. Centrifuge the lysate for 10 min at maximal speed in microcentrifuge
  5. Recover the above yeast extract for further analysis.
